You have to do this in xx-XX.com_ckforms.ini language file for admin, where xx stands for the language you are using.

In my case (using the plugin) I do not have this file!

Where can I change or remove the word "required"?

If you do not have this file then you didn't install your language file.
If your language is not translated yet, feel free to do this, or download the english language file and install. There you can change the required titles.

I know that, already installed en translation via joomfish. Now I am wondering where could I change the "Required *" string. Can I translate it through joomfish or I have to do it somewhere manually and in which file?

Whatever language you use - here is an example for german language:
C:\xampp\htdocs\joomla\administrator\language\de-DE where joomla stands for the site name.
